Buds of x Malosorbus florentina (Zucc.) Browicz (Rosaceae) collected in March, May and June 2004 were tested as regards their ability to install in vitro cultures. The surface sterilization of buds with 20 or 25% commercial bleach solution for 10 or 15 min was found more effective in reducing infections' percentage than the sterilization with 10 or 15% commercial bleach solution for 10 min. The removal of external scales along with bark reduced the infection of explants. Buds collected in March were cultured on solid MS with 0.7 or 1 mg/L BA with or without 0.1 mg/L IBA. Buds of May were cultured on MS with 1 mg/L BA with or without 0.1 mg/L IBA or NAA and buds of June on MS with 1 mg/L BA with or without 0.1 mg/L IBA or with 1 mg/L kinetin with or without 0.1 mg/L IAA or with 0.05 or 0.2 mg/L TDZ. Buds collected in March and June sprouted at low percentage (lower for June's buds) on media with BA and IBA, while buds collected in May formed only callus from which the regeneration of shoots was not possible. The browning and necrosis of explants were severe, probably because of the release of phenolics into the medium. The microshoots from buds collected in March were subcultured five times on MS with 1 mg/L BA and 0.1 mg/L IBA giving high shoot multiplication rates in contrast to the microshoots from buds collected in June that showed low multiplication rates. |
